An earlier pilot trial found a faster result on one attention task an hour after a single lion's mane dose, but that was one measure among many[2]. Put simply: you shouldn't expect to feel a gummy "kick in."

What might you notice over weeks?

The trials that found something positive were longer. In a 16-week study, older adults with mild cognitive impairment who took about 3 grams a day of dried lion's mane scored better on cognitive tests than those on placebo, and the difference faded after they stopped[3]. In the 28-day trial in young adults, the lion's mane group showed a trend toward lower perceived stress, though it wasn't a clear, statistically significant effect[2].

So if people notice anything, it's more likely to be a quiet sense of steadiness over weeks than a moment-to-moment change.
